External AW water plumbing

So I'm switching to an inline external water pump for my air to water this year( over a bilge type) and kind of looking for ideas on how to run the lines? Does the pump have to be at the lowest part of the system? How much pressure is lost if having to make it go vertical to reach intercooler? Any help would be appreciated.

Most all pumps need to be self priming. I'm a boat mechanic an know which ones are an not, if that helps. What pump?

I looked up the Mezier pump, most defiantly needs to be low mounted an force primed.
